I have a question, since XNA has no internal design tool for level editing, I wanted to know whether or not I can use Blend Expression 4 or Flash CS5 to design levels?

You can use whatever you like for building content, you just need to write a content loader to get it into your game, and then interpret that data to create your level. Take a look at this article for help.
