Seprating textures

Ok I can load and draw a texture from file but I do not know how to seperate it into tiles for my 2d game. I guess if someone knew a good toturial on D3DXSprite it would solve my problem. 2cd problem I save the texture as .png and am useing "paint shop pro 7" but when i texture the tiles they do not use transparncy. any help on either of these would be apprciated.
