Texture-collision_detection related question!

How can I scan through a rect on which a texture has been applied and get for each pixel the correspoding color and the alpha value so I will be able to store them in an array and use it for collision detection? If anyone knows a way to make collision detection using alpha channels or a good tutorial,please let me know.

Lock the texture.
