2D Sprites
You can used To Photoshop to make those sprite in my opinion is not the best tool for it but it can be used to do very detailed sprites and stuff
I think most of the time, sprites start as 3d models. Images are taken of them at different animations and from different angles. Animation will look more realistic, and lighting can be applied to the model which will make the sprites to appear turely 3D. Correct me if im wrong, but 3ds max and a number of other modeling packages have tools for doing just this. Edit: IMO, it would look really awsome if you applied cell shading to the mesh!
