I'm going to be developing a series of functions that I'd like to use from Python so my questions is should I write these functions C, C++ or use something like Cython to write the functions? The reason why I'm not doing these functions in Python to begin with is that they are a mix of CUDA and other functions with data Python can not touch.

Choose the one you're most comfortable with for developing the functionality you want. Python doesn't care what language its extensions are written in; just that the extension matches the import format, and once you have your functions written and compiled you can use whatever binding system you want, be it boost::python, SWIG, Cython or by hand.
