Making a fighting game is possible but you might want to start with something simpler like Pong, etc.

If you've chosen your language, you've already made the first step. Afterward, I might recommend reading some books. There are pretty much some for almost every language or tool. If you want something a bit cheaper than books, there are tutorials, which you can (usually) get for free.

Good advice:

AND work through every example, changing one or two things to ensure you're learned the information, not just copied and compiled the code.

I have no idea what your are asking, especially what you even mean by 'manipulation'. It's not a word normally used without any additional context in graphics programming.

There are no fundamental differences regarding what you can do with OpenGL or Direct3D. The APIs work a bit differently and people like one or the other for various reasons, but the results a skilled programmer gets out of either are identical for all practical purposes. If we talk about restrictions, then Direct3D is more restricted because you might have to install a newer operating system to get access to the newest while driver update is enough for OpenGL thanks to extensions. But even that is usually irrelevant unless we get into a new 70%-of-users-are-stuck-on-DirectX-9 situation.
