16bit colour key in direct draw

I know I know, ANOTHER question about using 16/32 bit and DirectDraw! Sorry, but this is my last one, honest! I am loading my background and sprites and they display fine, apart from my all my sprites still have a black background. In 8 bit, I set the colour key (or color key) just after i create the surface to hold the sprite image, specifying what palette entry to use (0 for black). How do I do this in 16/32 bit? I currently have both the low and high value for the colour key set to _RGB16BIT565 (0,0,0) but that black background still shows. If anyone can tell me how and when I should set the colour key I would appreciate a reply, Cheers Wizzard
